CCMV Delta 26 coat protein with N-terminal K-coil
CCMV is an abriviation of the Cowpea Chlorotic Mottle Virus, one of the most studied viruses in producing Virus Like Particles (VLPs). This part is the coding sequence of the coat protein of the CCMV virus, with 26 AA deleted on the N terminus, and a leucine zipper (K-coil) attached to the N terminal end with a flexible linker. When expressed in Escherichia coli, these coat proteins can be harvested and be assembled in vitro using the [http://2012.igem.org/Team:Wageningen_UR/Protocol/StartupCCMV| Wageningen_UR iGEM 2012 protocols]. With this part we tried to get an leucine zipper to the exterior of the VLP, obtaining a tool to achieve an universal platform where we use this anchor like system as a [http://2012.igem.org/Team:Wageningen_UR/Coil_system#The_Plug_and_Apply_.28PnA.29_System| Plug 'N Apply System].
Usage and Biology
This part is provided for the iGEM community so it can be used and integrated in an expression biobrick (e.g. BBa_K883001). This can be used to produce CCMV VLPs for their own use, including but not limited to packaging, making fusion proteins to the C terminal and coating the VLP with ligands and epitopes equiped with an opposite charged E-coil.
