This is a plasmid backbone that was designed to have its copy number (number of copies of the plasmid in one cell) controlled by the presence or absence of the inducer molecule Isopropyl β-D-1-thiogalactopyranoside (IPTG). This copy number in turn corresponds to the expression level of the genes on the plasmid.
This biobrick has been tested in an experiment looking at plasmid loss from a culture of cells that are not growing under selection. After two tubes of growth medium (identical with the exception of exactly one tube having IPTG present) had been inoculated with TOP10 containing this plasmid, they were left to grow at 37(C). After 24 hours these cultures were tested for the number of cells that would produce a colony when plated onto media with both selection as well as IPTG. The result was that the culture containing IPTG produced approximately 50 times as many colonies as the culture without IPTG.
