pLac-RBS-GFP-T-pTet-LacI
This mechanism was originally designed to simulate one state of the toggle switch BBa_K876001. It was used to test the efficacy of IPTG binding to LacI and LacI binding to LacO.
In addition to the previously mentioned functions, this part can be used as a biological sensor for IPTG. Under standard conditions, LacI is constitutively expressed and binds to pLac, preventing the transcription of GFP. As IPTG is added to the system, LacI is bound and inhibited, enabling the generation of GFP. Therefore, an increase in GFP can serve as an indicator for the increase of IPTG.
