Toggle Mechanism
A toggle mechanism that utilises the effects of TetR and LacI on pTet and pLac to lock DNA transcription to a specific state.
When pLac is running, TetR and RFP will be generated. TetR binds to pTet and prevents the transcription of LacI and GFP. The RFP serves as an indicator that TetR is being generated. When ATc is added to the system, TetR is bound by the ATc and can no longer prevent the transcription of LacI and GFP. As LacI accumulates, it binds to pLac and prevents any further transcription of TetR or RFP. At this point, the E. coli will lock into a green fluorescent state as the transcription of TetR and RFP drops.
On the contrary, if IPTG is added to the system, LacI will be bound and unable to inhibit the pLac promoter. Therefore, TetR and RFP will be constitutively expressed and pTet will be inhibited. The E. coli should lock into a red fluorescent state.
