TCC Negative control
This translational coupling cassette uses a version of UW-Madison's codon-optimized LIMS1 gene, (BBa_K762100,) with a stop codon in the middle of the sequence. When this gene is cloned upstream of the translational coupling cassette, the stop codon will prevent the helicase activity of the ribosome from breaking the mRNA hairpin, preventing translation of the red fluorescent protein (RFP) reporter gene. Negative control target gene for the translational coupling cassette made by UW-Madison 2012's team.
