ODR7:ADBR2:UNC-54 (for use in C. elegans)
Composite part designed to modify chemotaxis of C. elegans.
Characterization
Part was characterized through the use of a chemotaxis assay.
Agar plates were divided into four quadrants, 2 areas corresponding to 'target' and 2 'control' sections. In the target sections, differing concentrations of phenol were placed while control section used distilled water. Sodium azide was used as a paralytic in each quadrant to 'freeze' worms once they chemotaxed to that quadrant. Target chemical used included naphthalene, pond tailings sample, bitumen sample, and 4-chlorobenzoic acid.
Worms were placed at the centre of the agar plate (inside circle) and were allowed to chemotax for one hour. After one hour, worms were counted in each quadrant. Any worm contained in the circular zone approximately 0.5mm around centre of agar plate was not counted as either in a control or target quadrant. An example agar plate is shown below.
To determine quantitative measurement, the chemotactic index was determined. The formula for this index is shown below:
Result
The results of the chemotaxis assay for the ODR-7:ADBR2:UNC-54 part are shown below.
Preliminary results for a pond tailings mixture also appear to indicate a strong chemotactic index from the transgenic worms, however this data has not been characterized. Comprehensive testing and characterization of other chemicals (involving statistical analysis) will take place following iGEM Americas Jamboree to support these findings.
