Gli1-ADC HivC-AAR Construct

This part encodes Gli1-Aldehyde Decarbonylase and HivC-Acyl-ACP Reductase Zn-finger fusion proteins under the control of a lac-induced promoter. These co-localize when co-expressed with Slovenia's 2010 DNA Program, and produce alkanes.

Usage and Biology

Our attempt at zinc finger localization resulted in an alkane production of approximately 7 mg/L, which is significantly less than our delocalized PetroBrick standard, which produced around 36 mg alkanes per liter. We chose to integrate the use of DNA localization via zinc finger proteins to colocalize ADC and AAR to increase system flux.
Our gas chromatography-mass spectroscopy (GC-MS) data shows that the alkane yields from zinc finger were low. For more info, refer to 2011 UW iGEM Team wiki
