LacI-KRAB Repressor MammoBlock
This part encodes a repressor protein that binds tightly to the lactose operator originating from Escherichia coli. This protein is a fusion between the LacI repressor and the Kruppel-associated box (KRAB), a domain previously shown to strongly repress transcription from affected promoters via chromatin silencing. This repressor has been codon-optimized for mammalian expression and is capable of exerting very strong repression on promoters with internal lactose operator sites. Because LacI-KRAB-mediated repression can last for days due to its chromatin remodeling activity, LacI-KRAB is recommended for situations in which a change in transcriptional activity must be stable for extended periods of time, such as a permanent switch.
