This part is a pathway combination consisting of ptb, buk, adhE2, and ATF1, which can degrade butyrate, a four-carbon fatty acid, to ethanol and acetate esters. ptb is a phosphotransbutyrylase from Clostridium acetobutylicum, which can convert butyryl-CoA and phosphate to butyryl phosphate and CoA. buk is a butyrate kinase from Clostridium acetobutylicum, which can convert butyryl phosphate and ADP to butyrate and ATP. adhE2 is an aldehyde/alcohol dehydrogenase from Clostridium acetobutylicum, which can catalyze the reversible conversion of butyraldehyde and NADH to butanol and NAD+. ATF1 is an alcohol acetyltransferase from Saccharomyces cerevisiae, which can transfer the acetyl group from acetyl-CoA to alcohols to produce acetate esters. The part also contains a strong bacterial ribosome binding site (RBS) (BBa_B0034) before each gene to ensure efficient translation. A double terminator (BBa_B0015) is added at the end of the part to prevent read-through transcription and ensure the stability of the mRNA.
