Part:BBa_K4773015
Lac Promoter+RBS+cotA+RBS+QsrR+RBS+catA+Term.
This part is a gene combination consisting of cotA, QsrR, and catA, which can degrade benzo[a]pyrene, a carcinogenic polycyclic aromatic hydrocarbon, to less toxic compounds. cotA is a multicopper oxidase from Bacillus pumilus WH4, which can oxidize benzo[a]pyrene to quinones and hydroquinones. QsrR is a transcriptional repressor from Staphylococcus aureus, which can sense and respond to quinone stress by regulating the expression of genes involved in quinone detoxification. catA is a catalase from Escherichia coli, which can decompose hydrogen peroxide, a byproduct of cotA activity, to water and oxygen. The part also contains a strong bacterial ribosome binding site (RBS) (BBa_B0034) before each gene to ensure efficient translation. A double terminator (BBa_B0015) is added at the end of the part to prevent read-through transcription and ensure the stability of the mRNA.
