Part:BBa_K4773012
Lac Promoter+RBS+SQR+RBS+SDO+RBS+AprBA+RBS+SAT+Term
This part is a pathway combination consisting of SQR, SDO, AprBA, and SAT, which can degrade hydrogen sulfide (H2S), a toxic and malodorous gas, to sulfate (SO42-) and adenosine 5’-phosphosulfate (APS) in aerobic bacteria. SQR is a sulfide:quinone oxidoreductase from Acidithiobacillus thiooxidans, which can oxidize H2S to elemental sulfur (S0) and transfer electrons to quinone. SDO is a sulfur dioxygenase from Aquifex aeolicus, which can convert S0 to sulfite (SO32-) using molecular oxygen. AprBA is a dissimilatory adenosine 5’-phosphosulfate reductase from Desulfovibrio vulgaris, which can reduce APS to sulfite and AMP in sulfate-reducing bacteria. SAT is a sulfate adenylyl transferase from Escherichia coli, which can synthesize APS from sulfite and ATP. The part also contains a strong bacterial ribosome binding site (RBS) (BBa_B0034) before each gene to ensure efficient translation. A double terminator (BBa_B0015) is added at the end of the part to prevent read-through transcription and ensure the stability of the mRNA.
