This part is derived from the genomic sequence of Pseudomonas putida S16, a nicotine-degrading bacterium isolated from tobacco waste. And was cloned into the pUSP vector with the BBa_B0034 RBS.


This part contains a strong bacterial ribosome binding site (RBS) (BBa_B0034) followed by the coding sequence of NicB, a nicotine-degrading enzyme from Pseudomonas putida S16. NicB catalyzes the hydrolysis of pseudooxynicotine to 6-hydroxy-3-succinoylpyridine, which is further degraded by other enzymes in the nicotine catabolic pathway. This part can be used to engineer bacteria that can degrade nicotine in tobacco products or in the environment. NicB has a molecular weight of about 50 kDa and requires Mg2+ as a cofactor. NicB has an optimal pH of 7.5 and an optimal temperature of 30°C. NicB is active against other pyrrolidine derivatives, such as nornicotine and anatabine.
