Rigid Linker
This part is a rigid and stable alpha helix-forming linker that allows for a stiff space between protein structures within a recombinant protein. In many circumstances, rigid linkers are able to separate the functional domains or different proteins within a fusion protein (Chen et al. 2013). This property enabled the 2023 UBC-Okanagan team to fuse VP3 (a virus protein) and CotB (a surface membrane protein) without disfiguring any of the proteins’ structures.
The linker is 20 amino acids long. Glycines were added at the 5’ and 3’ ends of the sequence.
The sequence has been codon optimized by the 2023 UBC-Okanagan team for expression in the gram-positive model organism, Bacillus Subtilis.
