tph operon from Comamonas testosteroni
BBa_K4728012 is a composite part made up of five genes with six ribosome binding sites. These genes are tphC, tphA2, tphA3, tphB, and tphA1. TphC is a periplasmic TPA binding receptor protein. The periplasmic TPA binding receptor is believed to be involved in TPA uptake into the cell. This TPA is later degraded into DCD by TPADO. tphA2, tphA3, tphB, and tphA1, code for the large subunit of TPADO, small subunit of TPADO, a DCD dehydrogenase, and the reductase component of TPADO, respectively. The large subunit, small subunit, and reductase component of TPADO, along with the large subunit from P-1-B1 will degrade TPA into DCD. The DCD dehydrogenase will further degrade DCD into PCA. These two genes both come from a gene cluster known as tphRICIA2IA3IBIA1I isolated from Comanomas sp. strain E6.
The RBS to separate and flank the multiple CDS is the version of B0030 (BBa_J428032) provided through the iGEM 2023 distribution kit, which has a 6bp scar.
