FAST-PETase (Engineered PET hydrolase)
An improved version of IsPETase from I. sakaiensis (BBa_K1921000), with mutations N233K/R224Q/S121E/D186H/R280A [1]. The variant shows improved PET degradation activity over previous IsPETase and LCC variants at a wide range of temperatures from 30°C to 50°C. At sufficiently high concentrations FAST-PETase can fully degrade untreated consumer PET containers of moderate crystallinity.
This sequence was used in part BBa_K4701300, which is designed to produce extracellular FAST-PETase our of E. coli hosts.
