gRNA-(PAM/TTTA(+))

The first step is to select a guide sequence that matches the specific DNA sequence of the target gene, and then synthesize it as a gRNA with the designed guide sequence and RNA backbone. gRNA sequences are then integrated into expression vectors using primers. In this way, a plasmid with a specific gRNA is constructed. Afterwards, the gRNA was transferred into Corynebacterium glutamicum by transformation to verify the selection of the gRNA by observing the number of colonies on the final plate and the editing efficiency.
