Lac Promoter
Our contribution is that our work shows that a derepressed lac promoter can be successfully used in Rhizobia and act as a promoter for the PstSCAB, pqq, and gcd genes. The Lac promoter is constitutively activated and is the promoter we used in our plasmid, pBBR1MCS-2. We chose this promoter because it is a well-documented promoter known for being a strong promoter and because it is induced by the presence of lactose. This part can be used in any biosafety level laboratory. We used IPTG as a lactose substitute for activation to test the functionality of our promoter in our plasmid. We verified the sequence of our promoter and tested the ability of the plasmid to pick up a DNA insert by performing a blue-white screening. The intact plasmid contains a functioning LacZ gene, so when we add Xgal and IPTG to the plasmid, it should turn blue. However, when the genes have been inserted into the plasmid and the LacZ gene has been disrupted, the plasmid should turn white.
