pstC codon optimized for Rhizobium tropici
pstC, along with pstA, form membrane integral proteins that form a membrane channel in the periplasm of a bacteria, allowing for transport of phosphate into the cell. It is important because it enables the uptake of phosphate by creating an ion channel in the cell membrane through which inorganic phosphate can pass. We designed this part based on research from literature, such as this paper https://bmcmicrobiol.biomedcentral.com/articles/10.1186/s12866-019-1445-3 which further documents the well known pstSCAB system which is known to increase phosphate uptake. We then codon optimized this part for rhizobia, our chassis of choice. We obtained this DNA from Integrated DNA Technologies. We did not encounter any problems with obtaining and using this part. This part can be use din biosafety level laboratory. We recommend that this part be tested in rhizobia, since it is codon optimized for rhizobia compatibility, but it can be modified to work in any bacterial strain. To test this part, we ran a pcr with primers specific to this part, and ran a gel. Since the gel yielded positive results, we know that this part was successfully expressed. During the gel electrophoresis we used a standard negative control to compare the results of the primers.
