pCA24N-csgA-spytag
This section is used to express the fusion protein CsgA-Spytag as one of the components of our project product in vivo functional biofilm. We first carried out crystal violet and Congo red experiments. Please refer to experiment for detailed protocol. https://2023.igem.wiki/neu-china/experiment
Fig.1 Results of Crystal Violet Experiment
Fig.2 Results of Congo Red Experiment
The comprehensive experimental results showed that the addition of Spytag had little effect on the film-forming ability of chassis bacteria, while Mfp5 had great influence on the secretion of CsgA due to its large molecular weight, which led to a significant decrease in the film-forming capacity of chassis bacteria. In the follow-up, we tested the adhesion of biofilm, the anticorrosion ability of biofilm and the biomineralization test. For more information, see BBa_K4635006
