ccdB Toxic Protein Involved in the Construction of Suicide Switch.
In order to prevent the expression of csgA gene in E.coli genome from affecting the expression of csgA-spytag gene we constructed, the wild csgA gene in E. coli genome was knocked out by λ-RED gene recombination technology, and the toxic protein ccdB gene regulated by arabinose inducible promoter was designed to replace csgA gene in E. coli genome.
At the same time, for the sake of biosafety, it is involved in the construction of a suicide switch for Bacillus subtilis. We added an arabinose induced promoter upstream to facilitate the induction of death of Bacillus subtilis by adding arabinose when needed.
