XbaI-stgRNA-1-SpeI-BamHI-selfRecombination
XbaI-stgRNA-1-SpeI-BamHI-selfRecombination is the cassette for illumination form of self-targeting guide RNA 1. This part can be transcripted as a piece of guide RNA to target its own fragment and achieve a knock-out. Then Lambda system can implement the repair of the knockout fragment later. If the repair has been done correctly, the fragment can be connected and the green fluorescent protein gene can be expressed. Then we will observe green fluorescence. In other words, we can identify whether the fragment has been knocked out and repaired.
This is the first process of our multi-knock out-repair system. In the next step, we will try to exchange the detection protein with the second stgRNA fragment part to continue the knock out-repair process.
