code_ hsa-let-7b-5p (BBa_K461240) is a 23-nucleotide single-stranded DNA sequence that is a first-order weighted strand of the target hsa-let-7b-5p, designed by NUPACK from the target sequence. The primary weight probe has its partial complementary sequence on it, positioned after the target (5' to 3'). In order to realize its target hsa-let-7b-5p weight number size 8, we use the cascade method to transform 8 into 4*2, so the first-level weight probe of hsa-let-7b-5p combines 4 chains of this weight. The 18 nt at the 5' end of the weighted strand is paired with the weighted probe, and the last 5 nt is not paired with the weighted probe. The presence of this free 5 nt sequence prevents the DNA polymerase from combining with the weighted probe between the weighted strands and initiating a polymerase-mediated strand displacement reaction, resulting in the number of weighted strands displaced, released and less than the corresponding weights of the target.
