hsa-let-7b-5p _S2(BBa_K4612135) is a 64-nucleotide single-stranded DNA sequence that serves as a secondary weight probe for the target hsa-let-7b-5p. The sequence at its 3' end is designed to be complementary to the primary weight strand so that it can bind specifically to the primary weight strand on the primary weight probe when it is released, thus accomplishing recognition of the input signal. the DNA polymerase polymerizes DNA in the 5'-3' direction starting from the 3' end of the primary weight strand. In order to realize its target hsa-let-7b-5p weight number size 8, we adopt the cascade method to convert 8 into 4*2, so there are also 2 secondary weight chains bound to this probe, and the secondary weight chains will be replaced and released by the newly synthesized daughter chains during the process of DNA polymerization by Bst DNA polymerase. Through this process, the signal of the target is quantitatively converted into the number of weighted strands, which facilitates subsequent calculations, and finally the results of the operation are converted into different fluorescence intensities, making the results simple and easy to read.
