hsa-let-7b-5p (BBa_K4612132) is a 99-nucleotide single-stranded DNA sequence that serves as a first-order weighted probe for the target hsa-let-7b-5p. The 3' end of the sequence is designed to be complementary to the target hsa-let-7b-5p so that it binds specifically to the target and thus recognizes the input signal. DNA polymerase polymerizes the DNA in the 5'-3' direction from the 3' end of the target hsa-let-7b-5p. In order to achieve the target hsa-let-7b-5p weight size 8, we use the cascade method to convert 8 into 4*2, so there are also 4 primary weight strands bound to the probe, and the primary weight strands will be replaced and released by newly synthesized daughter strands during DNA polymerization by Bst DNA polymerase, and the released primary weight strands will be used as target-specific recognition signals. The first-order weight strand will be used as the target to undergo polymerase-mediated strand displacement reaction again to realize the multiplication of weights. Through this process, the signal of the target is quantitatively converted into the number of weighted strands, which facilitates subsequent calculations, and finally the results of the operation are converted into different fluorescence intensities, making the results simple and easy to read.
