hsa-mir-375-3p _S (BBa_K4612128) is a 64-nucleotide single-stranded DNA sequence that serves as a weighted probe for the target hsa-mir-375-3p. The 3' end of the sequence is designed to be complementary to the target hsa-mir-375-3p so that it binds specifically to the target and thus recognizes the input signal.DNA polymerase polymerizes DNA in the 5'-3' direction from the 3' end of the target hsa-mir-375-3p. The DNA polymerase polymerizes DNA from the 3' end of the target hsa-mir-375-3p in the 5'-3' direction, and the probe binds a weighted strand that corresponds to the weight number of the target hsa-mir-375-3p (size 2), which is then replaced by a newly synthesized daughter strand that is released during the DNA polymerization process by the Bst DNA polymerase. Through this process, the signal of the target is quantitatively converted into the number of weighted strands, which facilitates subsequent calculations, and finally the results of the operation are converted into different fluorescence intensities, making the results simple and easy to read.
