LacI promoter
LacI promoter is based on the lactose manipulator model promoter and contains two protein binding sites. One site binds the LacI protein and the other site binds the cAMP activator protein (CAP, crp gene product), which is normally found in E. coli and is associated with cellular health and glucose availability.Lac promoter promotes transcription in the absence of the LacI protein and the CAP protein. In the presence of LacI protein and CAP protein, it inhibits transcription. In contrast, the addition of Isopropyl-beta-D-thiogalactopyranoside (IPTG, an isogalactose mimetic) inhibits the LacI protein, thereby inducing transcription.Lac promoter is widely used for prokaryotic expression because of the universality of its regulatory conditions, and can be further optimized for expression efficiency by controlling the concentration of IPTG.
