fuGFP-alanine

This is a blue-shifted variant of part BBa_K3814004 which has two base change in the fluorophore (hence the alanine nickname). It absorbs blue light and fluoresces green-yellow.

This part is a variant of fuGFP (Part:BBa K3814004) which was originally designed to create a GFP variant not protected by patents and able to facilitate any kind of research. Using PCR with degenerate primers we introduced the S66A mutation in the fluorophore to create fuGFPa. The variant has an excitation peak at 480 nm and emission peak at 505 nm (figure 1). We have also created a fusion protein of fuGFPa connected to a cellulose-binding domain which allows it to be immobilised on cellulose (Part:BBa K4488026).
