IK17 (anti-oxLDL) scFv antibody fragment
Overview
This sequence codes for a short chain variable fragment (scFv) derived from the anti-oxLDL IK17 antibody. Specifically, this scFv targets a protein structure called MDA-modified apo-B (apoB(MDA)) that is part of an oxLDL molecule. OxLDL stands for oxidized low density lipoprotein, and it is a biomarker for many inflammatory diseases such as atherosclerosis. As a result, antibodies like this one that target oxLDL could enable many diagnostic and therapeutic approaches for such inflammatory diseases.
This sequence codes for the following in sequential order: IK17 variable heavy chain, a (G4S)3 linker sequence, and IK17 variable light chain. The CDS is derived from the antibody IK17, which Tsimikas et al. found binds to apoB(MDA) (1). The variable heavy and light chain of IK17 are connected by a (G4S)3 linker sequence (2). This linker sequence was chosen because it was designed to enhance protein expression of recombinant antibodies. Note that the protein resulting from this sequence will only fold correctly in organisms capable of producing disulfide bonds, such as SHuffle E. coli.
