Human/mouse anti-apoB(MDA) (anti-oxLDL) IgG antibody - heavy chain
This sequence codes for the heavy chain of an anti-apoB(MDA) immunoglobulin G (IgG). ApoB(MDA) is a specific chemical structure found as a component of oxidized low density lipoprotein (oxLDL). OxLDL is a biomarker of many inflammatory diseases such as atherosclerosis, so antibodies able to target oxLDL could enable potential detection and therapeutic applications for such diseases. This part is best used when expressed as part of the same expression cassette with part BBa_K4477001, which encodes the light chain of this anti-oxLDL antibody. When expressed together, the light and heavy chains will associate to form a complete IgG. See part BBa_K4477011 for the composite part Team Virginia 2022 recommends for expressing both light and heavy chains together.
Like any antibody, Team Virginia 2022's hybrid anti-apoB(MDA) IgG requires disulfide bonds for it to be properly folded and exhibit the correct binding affinity. As a result, it is crucial that both the light and heavy chain CDSs be expressed in an organism capable of creating disulfide bonds while it expresses proteins, such as the SHuffle bacterial strain or most eukaryotic organisms. Note that many common bacterial strains do not have this capability! Team Virginia 2022 used SHuffle E. coli to express this hybrid anti-apoB(MDA) IgG.
