T7 gp0.7 (C-Terminal)

BBa_K4451007 encodes the C-terminal end of gp0.7 protein from T7 phage. This protein has been reported to interact with the RNA polymerase beta prime subunit and cause host transcription shutoff (Michalewicz and Nicholson, 1992).

A transcriptional inhibitor from T7 phage, which is able to cause growth rate suppression in E. coli.

Description

The Gp0.7 gene encodes a serine/threonine kinase which is responsible for the phosphorylation of RpoB and RpoC subunits of bacterial RNA polymerase, which increases the incidence of Rho-dependent termination of bacterial transcripts thus enacting a global transcriptional downregulation. Gp0.7 is utilised in tandem with the Gp2 gene (an inhibitor of RNA polymerase holoenzyme formation) in the T7 bacteriophage infection process (Michalewicz and Nicholson, 1992) (ES. Robertson and AW. Nicholson, 1990).
