pET28bBhp R is a primer sequence which is a short single-stranded DNA sequence that is used as a starting point for PCR amplification of a gene. Primers effectively "direct" DNA polymerases where to begin replication of DNA, because DNA polymerase can only add additional nucleotides to an existing strand of DNA since it cannot initiate DNA synthesis. This part will act as our reverse primer and has a length of 28 bp. It has the XhoI restriction enzyme recognition site and a 5’ overhang of 4
bases ‘GGTG’ for increasing the cleavage efficiency of the restriction enzyme XhoI. The primer will be needed for the PCR amplification of the Bhp gene which we will further use as a control for our experiment. It has 46% GC content with a Tm of 63 degrees Celsius.
