pET28bBhp F is a primer sequence which is a short single-stranded DNA sequence that is used as a starting point for PCR amplification of a gene. Primers effectively "direct" DNA polymerases where to begin replication of DNA, because DNA polymerase can only add additional nucleotides to an existing strand of DNA since it cannot initiate DNA synthesis. This part will act as our forward primer and has a length of 30 bp. It has the NcoI restriction enzyme recognition site and a 5’ overhang of 3
bases ‘ATA’ for increasing the cleavage efficiency of the restriction enzyme NcoI. The primer will be needed for the PCR amplification of the Bhp gene which will be further used as a negative control in our experiment. It has 33% GC content with a Tm of 58 degrees Celsius.
