pET15b FimH2 F is a primer sequence which is a short single-stranded DNA sequence that is used as a starting point for PCR amplification of a gene. Primers effectively "direct" DNA polymerases where to begin replication of DNA, because DNA polymerase can only add additional nucleotides to an existing strand of DNA, but cannot initiate replication. This part will act as our forward primer and has a length of29 bp . It has the Bam H1 restriction enzyme recognition site and a 5’ overhang of 4 bases ‘TCGA’ for increasing the cleavage efficiency of the Restriction enzyme BamHI. The primer will be needed for the PCR
amplification of our gene of interest Fim H2.It has 52% GC content with a Tm of 65 degrees Celsius.
