pET28b FimH 1R is a primer sequence which is a short single-stranded DNA sequence that is used as a starting point for PCR amplification of a gene. Primers effectively "direct" DNA polymerases where to begin replication of DNA, because DNA replication can only add additional nucleotides to an existing strand of DNA since it cannot initiate DNA synthesis. This part will act as our reverse primer and has a length of 30 bp. It has the Xho1 restriction enzyme recognition site and a 5’ overhang of 4 bases ‘GGTG’ for increasing the cleavage efficiency of the Restriction enzyme Xho1. The primer will be needed for the PCR amplification of our gene of interest Fim H 1. It has 53.3% GC content with a Tm of 80.1 degrees Celsius.
