pET28b FimH 1F is a primer sequence which is a short single-stranded DNA sequence that is used as a starting point for PCR amplification of a gene. Primers effectively "direct" DNA polymerases where to begin replication of DNA, because DNA polymerase can only add additional nucleotides to an existing strand of DNA, but cannot initiate replication. The DNA part will act as our forward primer and has a length of 32 bp. It has the Nco1 restriction enzyme recognition site and a 5’ overhang of 4 bases ‘ATATA’ for increasing the cleavage efficiency of the Restriction enzyme Nco1. The primer will be needed for the PCR amplification of our gene of interest Fim H1. It has 40.6% GC content with a Tm of 70.6 degrees Celsius. For allowing the inframe expression of Fim H1 an insertion of GG residues has been incorporated in the primer design just prior to the region complementary to the coding sequence.
