PmaeN, a malate inducible promoter for Bacillus subtilis
PmaeN is a native B. subtilis malate inducible promoter regulated by the MalKR Two-Component System to enable the usage of malate as a carbon source. Previous study (see doi link) has shown this promoter is not induced by other carbon sources such as fumarate, succinate or even gluocse - thus not under the control of carbon catabolite repression.
Using a luxABCDE reporter system, we demonstrate this promoter shows an increase in RLU/OD activity following subjecting cells to increasing concentrations of malate (L-maleic acid) to the point of promoter saturation using 0.5 mM malate.
