trigger-RNA sensor of igRNA (hsa_circ_0001785)
It is used as a sensing part of the RNA-interacting guide RNA (igRNA) in our CRISPR-Cas12 system to detect a unique sequence (trigger RNA) of the target circRNA hsa_circ_0001785. This part serves as a switch for the whole circuit. It contains three regions, VR1 region, loop region, and VR2 region. VR1 and VR2 regions complement to VR1’ and VR2’ regions (trigger RNA) of hsa_circ_0001785. Loop region is complementary to the guide of igRNA (hsa_circ_0001982) (BBa_K4409002). When hsa_circ_0001785 is present in the system, VR1 and VR2 regions combine with VR1’ and VR2’ sequences. Thus, loop region unblocks the guide of igRNA (BBa_K4409002) which can then bind with dsDNA(hsa_circ_0001982) (BBa_K4409008) and activate Cas enzyme to cleave DNAs.
