V5 Tag

The V5 epitope tag has the sequence GKPIPNPLLGLDST. The V5 tag is derived from a small epitope present on the P and V proteins of the paramyxovirus of simian virus 5 (SV5) corresponding to amino acids 95-108. Epitope tags can be fused to a protein of interest for detection in antibody-based methods such as Western blotting, immunofluorescence or flow cytometry. The availability of commercial, well-characterized antibodies makes using epitope tags like V5 a viable alternative to generating custom antibodies for the recombinant protein.

Experience

We utilised the V5 tag for our composite part (BBa_K4359008) and used anti-V5 antibodies (#13202, Cell Signaling Technologies) at a dilution of 1:1000 for Western blotting. We found minimal cross-reactivity with mammalian systems.
