CCP1_mod

CCP1 encodes mitochondrial cytochrome-c peroxidase, an enzyme that degrades reactive oxygen species in the mitochondria. In cells, it acts as a hydrogen peroxide sensor in mitochondria to produce a cellular response to oxidative stress (SGD:S000001774). We chose to utilize this inducible promoter for our system so we could overexpress GSH1 and ADP1 to increase glutathione synthesis and export only in response to elevated levels of oxidative stress in the environment. This conditional gene expression would increase the metabolic fitness of the yeast in our patch. We wanted to make this part compatible with Golden Gate assembly, so we removed the internal BsaI recognition site and cloned both the native promoter and this modified promoter to compare them to each other.
