Three-step inducible system with uvGFP
This plasmid contains one component out of three of the three-step inducible system. Here, the behaviour of the system with different lengths of the P1 spacer can be tested by measuring the uvGFP fluorescence. This construct consists of uvGFP coding sequence with a dSpyCas9 target site in between the promoters and coding sequence of this protein. Additionally, an introduction template for sgRNA spacers was introduced. This template contains two type IIS restriction sides, this way different length P1 spacers can be easily introduced and tested.
