Three-step inducible system with mRFP

This plasmid contains one component out of three of the three-step inducible system. Here the behaviour of the system with different lengths of the B1 spacer can be tested by measuring the mRFP fluorescence. This construct consists of mRFP coding sequence with a dSpyCas9 target site in between the promoters and coding sequence of this protein. Additionally, an introduction template for sgRNA spacers was introduced. This template contains two type IIS restriction sides, this way different length B1 spacers can be easily introduced and tested.
