SUMO protein tag

SUMO-tagged protein is a small ubiquitin-like modifier protein (Small ubiquitin-likemodifier), one of the important members of the ubiquitin-like peptide chain superfamily.

In terms of primary structure, SUMO has only 18% homology with ubiquitin, however, their tertiary structures and their biological functions are very similar. It was found that SUMO can act as a fusion tag and molecular chaperone for recombinant protein expression, which not only can further enhance the expression of fusion protein, but also has the function of anti-protease hydrolysis and promote the correct folding of target protein to improve the solubility of recombinant protein.

In addition, SUMO has an important application, which is to excise the tag protein intact and obtain the natural protein. This is because SUMO protein hydrolase (available from our company) recognizes the intact SUMO tag protein sequence and can efficiently excise SUMO from the fusion protein. After the SUMO is excised, the recombinant protein is obtained as the natural protein by affinity chromatography and removing the tag protein part. Therefore, SUMO tag is also commonly applied together with other tags as specific enzymatic cleavage hydrolysis sites.
