DNA template of mutated target RNA for LbuCas13a
This part can transcribe the mutated target RNA(Some mutation loci differ from BBa_K4223001, But belong to the same false positive test experiment) as the target sequence recognized by Cas13a1. Once Cas13a1 protein binds to sgRNA to form the Cas13a1-sgRNA complex, Cas13a1 in the complex can recognize the protospacer flanking sequence (PFS) of the target RNA and then change the conformation of Cas13a1 through base complementary pairing between sgRNA and target RNA after PFS recognition.Cas13a exhibits RNase activity and can cleave specific targeting single-directed RNAs (cis-cleavage activity). Cleavage of double-stranded DNA activates the ability of Cas13a1 protein to cleave non-specific ssDNA (trans cleavage activity).
In general, this is a segment of DNA that is transcribed to produce target RNA, and we can verify the identification of false positives and other phenomena by synthesizing this target RNA
