bFGF secretion system

Basic fibroblast growth factor (bFGF or FGF2) is a multifunctional growth factor. The BNUZH-China iGEM team 2022 designed a fusion protein consisting of secreted peptide (IL-2 signal sequence) and bFGF for the secretion of bFGF out of the cell. Therefore, we used our IL-2 signal sequence (BBa_K4211002) and fused it to the bFGF (BBa_K4211001). For protein purification and Western blot, we inserted a 6×His tag at the C-terminal of bFGF gene.


Figure 1: Construct design: The bFGF gene. The bFGF gene was inserted after the CMV Promoter, and the secrete peptide gene IL-2 Signal sequence (BBa_K4211002) was inserted in front of the bFGF gene.

Usage and Biology

We used bFGF (BBa_K4211001) and EGF (BBa_K4211000) to stimulate the growth of epidermal cells and fibroblasts, promoting the wound healing.
