p(TetR)-RBS-GFP-LVA-RBS-δ activator-LVA-terminator

The expression of EGFP and δ activator is regulated by the promoter p (tetR), which can be repressed by protein tetR. EGFP was used as a reporter gene. δ Activator is an activator from phage phiR73 that can act on a class of inducible promoters including pF promoters. In addition, in order to enable protein degradation in time, the downstream of EGFP and δ Activator was added with ssrA-LVA tag. SsrA tag is a 9~11-length amino acid sequence attached to the end of protein. Protein linked by SsrA tag can be degraded well by some combinations of protein (clpX, clpP and sspB).
