pSB1C3-FRET

This plasmid contains a FRET pair consisting of mTurquise2 and EYFP. Between these two dyes is the TEV-potease interface. With this system the presence or absence of the TEV Protease can be detected. If the protease is active, the linker containing the protease recognition site is cut. As a result, the distance between the two dyes increases and the energy transfer via the FRET system no longer works. You only see the fluorescence of mTurquise2. If the protease is inactive, the linker is not cut so the energy transfer can take place via the FRET system. It is therefore not the fluorescence of mTurquise2 but of EYFP that is to be seen.
