8XGTIIC motifs

8XGTIIC is 8 DNA motifs that contain TEA domains which are also called TEAD binding elements [1]. These domains are transcriptional binding sites that allow the TEAD transcription factors to bind to the DNA. YAP/TAZ proteins which are regulatory proteins in the Hippo pathway, translocate to the nucleus and regulate the TEAD proteins by binding to the GTIIC motifs [2].

Usage and Biology

The team of Technion 2022 used this part in a construct (BBa_K4179020) in order to quantify their manufactured metabolite-decursin. These motifs were found in the 8XGTIIC-Luciferase plasmid (#34615 AddGene) upstream to the luciferase gene. It is used as YAP/TAZ responsive Luciferase reporter. It has been proven that their manufactured metabolite affects the Hippo pathway by a cascade of phosphorylation that ends up regulating the TEAD proteins by binding to 8XGTIIC motifs.
