sgRNA

sgRNA guide dCas9 proteins to specific genomic locations through the principle of base-complementary pairing.The yeast CRISPRi webtool30 and CRISPR-ERA58 were used to design the sgRNAs, using as a criteria ATAC-seq values around 1 and nucleosome-free areas, to ensure accessibility to the target genes . Additionally, poly-N and presence of off-targets were avoided and GC content between 30 and 55 was preferred.
