An intrinsic terminator locating in the plasmid pBE2 Upstream the MCS of pBE2, there is a promoter S6 to initiate the transcription. If the transcription can be terminated well, there could be a terminator downstream of the MCS. So we used a terminator prediction online tool iTerm-PseKNC to analyze the sequence near MCS (http://lin-group.cn/server/iTerm-PseKNC/home.php). The analytic results show that there are terminators on both sides of MCS. With such a terminator, transcription of our regulatory protein XylR can be commonly stopped, and so that the promoter Pxyl can be repressed by XylR as the literature said.
