lactose regulated bla, RBS mutant

This device will not express b-lactamase regardless of environmental lactose concentration. The lacI biobrick contains a mutation in the RBS of bla that prevents the ribosome from binding. Specifically, the RBS was changed from AGGAAA to CCCTTT.

The mutation's function is observed using the test below. Since b-lactamase will not be made into a functional protein, no color change is expected when a culture is exposed to nitrocefin (b-lactamase chromogenic substrate).
